Association between NLRP3 inflammasome and periprocedural myocardial injury following elective PCI

چکیده

BackgroundPeriprocedural myocardial injury (PMI) is a common complication of percutaneous coronary intervention (PCI) associated with poor prognosis. Inflammation has been demonstrated to exert crucial role in PMI. However, how the inflammation initiated or sustained PMI remains elusive.MethodsRNA-seq peripheral blood mononuclear cells (PBMCs) from 3 Non-PMI and 6 patients was performed subsequent bioinformatics analysis. RNA-seq results were verified patient cohort. We also established microembolization (CME) mice model mimic The activity caspase-1 PBMCs detected by flow cytometry. levels interleukin (IL)-1β, IL-18 cardiac troponin plasma measured enzyme-linked immunosorbent assay.ResultsWe identified total 901 differentially expressed genes (DEGs) between patients. These DEGs participated several inflammation-related processes. NOD-like receptor signaling pathway significantly enriched All key composed NLRP3 inflammasome, including NLRP3, PYCARD, CASP1 IL1B, upregulated activation inflammasome then increased PBMCs, elevated IL-1β Spearman analysis confirmed tight correlations activity, IL-1β, T level. In addition, enhanced CME mice.ConclusionsWe discovered that involved PMI, thus providing evidence supporting therapeutic value inflammasome-targeted strategies

Association of chronic kidney disease with periprocedural myocardial injury after elective stent implantation

Coronary artery disease (CAD) is the leading cause of mortality in patients with chronic kidney disease (CKD). Patients with CKD who undergo percutaneous coronary intervention (PCI) may have more ischemic events than patients without CKD. The aim of our study was to determine the incidence of periprocedural myocardial injury (PMI) after elective stent implantation in patients with CKD using the...

Intracoronary ST-segment shift soon after elective percutaneous coronary intervention accurately predicts periprocedural myocardial injury.

BACKGROUND Elevation of cardiac biomarkers after coronary angioplasty (percutaneous coronary intervention [PCI]) reflects periprocedural myocardial damage and is associated with adverse cardiac events. We assessed whether periprocedural myocardial damage that occurs despite successful PCI could be rapidly and easily identified by intracoronary ST-segment recording with the use of a catheter gui...

NLRP3 Inflammasome Activation-Mediated Pyroptosis Aggravates Myocardial Ischemia/Reperfusion Injury in Diabetic Rats

The reactive oxygen species- (ROS-) induced nod-like receptor protein-3 (NLRP3) inflammasome triggers sterile inflammatory responses and pyroptosis, which is a proinflammatory form of programmed cell death initiated by the activation of inflammatory caspases. NLRP3 inflammasome activation plays an important role in myocardial ischemia/reperfusion (MI/R) injury. Our present study investigated wh...
